Activity of the antimicrobial peptide P34 against bovine alphaherpesvirus type 1 Ciência Rural

ABSTRACT: Previous studies have demonstrated the antimicrobial activity of the peptide P34. In this study, the antiviral potential of P34 and the in vitro mechanism of action were investigated against bovine alphaherpesvirus type 1 (BoHV1). P34 exhibited low toxicity, a high selectivity index (22.9) and a percentage of inhibition of up to 100% in MDBK cells. Results from antiviral assays indicated that P34 did not interact with cell receptors, but it was able to inhibit the viral penetration immediately after pre-adsorption. In addition, BoHV1 growth curve in MDBK cells in the presence of P34 revealed a significant reduction in virus titer only 8h post-infection, also suggesting an important role at late stages of the replicative cycle. P34 is an antimicrobial peptide produced by a Bacillus sp. strain isolated from the intestinal contents of a fish in the Brazilian Amazon basin with reported antibacterial activity. The aim of this work was to evaluate the peptide P34 for its in vitro antiviral properties against canine adenovirus type 2 (CAV-2), canine coronavirus (CCoV), canine distemper virus (CDV), canine parvovirus type 2 (CPV-2), equine arteritis virus (EAV), equine influenza virus (EIV), feline calicivirus (FCV) and feline herpesvirus type 1 (FHV-1). Complete genome sequence of acute viral necrosis virus associated with massive mortality outbreaks in the Chinese scallop, Chlamys farreri ArchiMer

Acute viral necrosis virus (AVNV) is the causative agent of a serious disease resulting in high mortality in cultured Chinese scallops, Chlamys farreri. We have sequenced and analyzed the complete genome of AVNV. The AVNV genome is a linear, double-stranded DNA molecule of 210,993 bp with a nucleotide composition of 38.5% G + C. A total of 123 open reading frames were predicted to encode functional proteins, ranging from 41 to 1,878 amino acid residues. The DNA sequence of AVNV is 97% identical to that of ostreid herpesvirus 1 (OsHV-1), and the amino acid sequences of the encoded proteins of these two viruses are 94-100% identical. Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

Detection of oyster herpesvirus DNA and proteins in asymptomatic Crassostrea gigas adults ArchiMer

Since 1972, several herpes-like virus infections have been reported among different bivalve species around the world. Most of these reports involved larvae or juveniles presenting high mortalities. A distribuição de anticorpos neutralizantes para o herpesvirus bovino 1 (HVB 1) foi estudada em quatro faixas etárias de bovinos, em 21 rebanhos de leite e de corte. Os resultados da sorologia foram analisados e relacionados com as respostas de questionários aplicados aos responsáveis pelos rebanhos. As taxas de freqüência de anticorpos neutralizantes para o HVB 1 foram comparadas segundo a aptidão e a faixa etária. Fatores como tipo de manejo e idade dos animais influenciaram na distribuição de anticorpos para o HVB 1.

Effects of temperature on herpes-like virus detection among hatchery-reared larval Pacific oyster Crassostrea gigas ArchiMer

This paper describes effects of temperature on herpes-like virus detection in Pacific oyster Crassostrea gigas larvae held at different temperatures. Intranuclear, intracytoplasmic and extracellular viral particles were observed in velum and mantle connective tissues of oyster larvae reared at 25-26°C. In larvae held at 22-23OC, although nuclear lesions were observed, the presence of viral particles was not detected. Results were obtained for oyster larvae with 4 different parental origins. Herpesvirus infection was found in 3 of the 4 groups of oyster larvae held In the same conditions at the higher temperature.

Evidence for interspecies transmission of oyster herpesvirus in marine bivalves ArchiMer

Since 1991, numerous herpesvirus infections associated with high mortality have been reported around the world in various marine bivalve species. In order to determine whether these infections are due to ostreid herpesvirus-l (OsHV1), a previously characterized pathogen of the Japanese oyster (Crassostrea gigas), PCR analysis was carried out on 30 samples of larvae collected from four bivalve species (C. gigas, Ostrea edulis, Ruditapes decussatus and Ruditapes philippinarum), most exhibiting mortality prior to collection. French scallops: A new host for ostreid herpesvirus-1 ArchiMer

Sporadic high mortalities were reported among larval French scallops (Pecten maximus). Electron microscopy of moribund larvae revealed particles with the characteristics of a herpesvirus in association with cellular lesions. PCR and DNA sequencing showed that the virus is a variant of ostreid herpesvirus-1 that has already been described in clams and oysters. This is the first description of a herpesvirus infection of a scallop species. The virus was transmitted successfully from an extract of infected scallop larvae to uninfected scallop or oyster (Crassostrea, gigas) larvae, demonstrating that it is able to infect both species. In 1972, Farley reported the presence of herpes-type viral particles in adult American oysters (Crassostrea virginica) for the first time. Since then, this pathogen has been detected in various marine bivalve species, usually associated with massive mortality events. To date, herpes-type viruses have been detected in the following species: C gigas, Ostrea edulis, O. angasi, Tiostrea chilensis, Ruditapes decussatus, R. philippinarum and most recently, Pecten maximus.

Inhibition of DNA virus: Herpes-1 (HSV-1) in cellular culture replication, through an antioxidant treatment extracted from rosemary spice BJPS

This work aimed to evaluate antiviral properties in antioxidants from spices. Phenolic compounds extracted from rosemary (Rosmarinus officinallis, L) by hot water, had their antioxidant activity determined by spectrophotometry using β carotene/linoleic acid system. The rosemary extract was evaluated by antiviral assay of Herpes Virus type-1 (HSV-1) replication in VERO cells, in the presence or absence of the spice. 10,000 TCID50/mL of the HSV-1 was kept for 3 h at 4º C, with 300 ppm of rosemary extract, and 100 ppm of butyl hydroxyl toluene (BHT).
